| Abstract: The present application relates to microfabricated columns having a sealed flow channel. In particular, the channel is sealed during dicing, thereby preventing debris from accumulating within channels. In use, the seal is ruptured mechanically to connect one or more fluidic connections, which deliver analytes to the flow channel. Additional details are provided for making and using sealed columns. |

| Advantage: The column keeps gas chromatography free of debris and contaminants during a dicing process and extended storage time for the column without fear of atmospheric contaminants entering the channels. The column allows structures to seal the channels and fluid connections, thus preventing water and dicing debris from entering the gas chromatography. |
| Novelty: The column (100) has a flow channel (120) arranged within a substrate (110). A proximal connector region (130) and a distal connector region (135) are in fluidic communication to the flow channel. A proximal seal (150) is arranged in the proximal connector region. A distal seal (155) is arranged in the distal connector region. The proximal seal and the distal seal comprise a dimension ranging from about 10 meter to 500 meter. The distal seal and proximal seal are configured to be mechanically ruptured. A lid is arranged beneath the substrate, where a base material is made of silicon or glass. |
