Abstract: Provided herein are single domain antibodies (sdAbs) configured for passing the blood brain barrier in a human or mammal brain. Methods of using such sdAbs are also described herein, such as methods of binding to brain cells, binding with and transporting other sdAbs with beneficial functionalities, binding to and transporting biochemical or pharmaceutical species with beneficial functionalities, and treating, diagnosing, or prophylactically treating a disease, malignancy, or condition.

Novelty: Single domain antibody construct comprising first single domain antibody with binding domain, is new; where binding domain comprises first polypeptide comprising first complementarity determining regions encoding 25 sequences of 7 amino acids (SEQ ID NOs: 1-25), given in the specification; second complementarity determining regions encoding 25 sequences of 8 amino acids (SEQ ID NOs: 26-50), given in the specification; and third complementarity determining regions encoding 25 sequences of 9-15 amino acids (SEQ ID NOs: 51-75), given in the specification.
