| Novelty: Reaction mixture comprises: (i) target nucleic acid comprising regions, including a B3 region, B2 region, B1 region, F1c region, F2c region and F3c region from 5' to 3' direction; (ii) first amplicon having corresponding regions sufficiently complementary to regions in target nucleic acid; (iii) duplex comprising forward inner primer comprising first nucleic acid sequence and further comprising second nucleic acid sequence and quench probe comprising third nucleic acid sequence having sufficient complementarity to first portion of forward inner primer and further comprising quencher label operably linked to third nucleic acid sequence; (iv) backward inner primer comprising fourth nucleic acid sequence having sufficient complementarity to B2c region of first amplicon; (v) pair of outer primers comprising forward primer comprising sixth nucleic acid sequence having F3 region and backward primer comprising seventh nucleic acid having B3 region; and (vi) pair of loop primers. |
