Microfluidic platform for multiplexed detection in single cells and methods thereof
DWPI Title: Performing multiplexed analysis in microfluidic device, by loading and capturing test sample portion, treating sample portion with reagent, incubating, amplifying and detaching amplified portion, and performing on-chip flow cytometry assay
Abstract: The present invention relates to a microfluidic device and platform configured to conduct multiplexed analysis within the device. In particular, the device allows multiple targets to be detected on a single-cell level. Also provided are methods of performing multiplexed analyses to detect one or more target nucleic acids, proteins, and post-translational modifications.
Use: The method is useful for performing multiplexed analysis in a microfluidic device (claimed) to detect one or more target nucleic acids, proteins, and post-translational modifications in single cells.
Advantage: The method allows rapid multiplexed detection of desired protein and nucleic acid targets, and the amount of reagent required to detect nucleic acids, proteins, and post-translational modifications is reduced by 95%.
Novelty: Method for performing multiplexed analysis in a microfluidic device, involves (i) loading a test sample portion into a main port of the device, (ii) capturing the test sample portion within each assay chamber, thus providing a captured sample portion, (iii) treating the captured sample portion with a first fixative reagent in each assay chamber, thus providing a fixed sample portion, (iv) incubating the fixed sample portion with a first protein label in each assay chamber, (v) incubating the labeled sample portion with a second protein label in each assay chamber, (vi) incubating the multi-labeled sample portion with a first nucleic acid label, (vii) amplifying a signal of the first nucleic acid label, thus providing an amplified sample portion, (viii) detaching the amplified sample portion, thus providing a detached sample portion, and (ix) performing an on-chip flow cytometry assay of the detached sample portion.
