Promoters and uses thereof

DWPI Title: New phenolic-inducible synthetic promoter comprising spacer region that comprises first element, second element and third element comprising specified nucleic acid sequences, used e.g. to treat lignin
Abstract: The present invention relates to synthetic promoters, as well as expression cassettes, and recombinant scaffolds, and methods thereof. In particular embodiments, the synthetic promoter is inducible by one or more chemical compounds present during lignin depolymerization.
Use: The phenolic-inducible synthetic promoter is useful: in an expression cassette; as a recombinant gene; as a recombinant vector; in an isolated host cell; and for treating lignin or its derivative (all claimed).
Advantage: The synthetic promoter: is inducible by at least one chemical compounds present during lignin depolymerization; can provide a dynamic feedback system during lignin breakdown; can provide enhanced protein expression of a nucleic acid that is operably linked to the promoter, hence provides efficient breakdown of complex compounds e.g. polymers (e.g. lignin); facilitates the transcription of a gene and/or binding of a polymerase to at least one promoter sites; offers lower operating costs by replacing expensive inducers with in situ inducers that are present or produced during lignin breakdown; and provides increased flexibility in operating conditions.
Novelty: Phenolic-inducible synthetic promoter comprising a spacer region, is new, where the spacer region comprises: (a) a first element comprising a nucleic acid sequence having at least 80% sequence identity to any one of a nucleic acid sequences of SEQ ID NOs: 22-25; (b) a second element comprising a nucleic acid sequence having at least 80% sequence identity to any one of a nucleic acid sequences of SEQ ID NOs: 26-33 (each comprising fully defined sequence of any one of 17-19 nucleotides as given in the specification); and (c) a third element comprising a nucleic acid sequence having at least 80% sequence identity to any one of a nucleic acid sequences of SEQ ID NO: 34-37, and the promoter comprising a nucleic acid sequence having at least 98% sequence identity to any one of a nucleic acid sequences of SEQ ID NOs: 1-9 (each comprising fully defined sequence of any one of 76-88 nucleotides as given in the specification).
